随着城市园林绿化事业的发展,对观叶植物引种的需求日益见长,随之对观叶树种的研究也逐渐被提到城市绿化的日程上来。紫叶矮樱为世界著名的观叶树种,国内关于紫叶矮樱组织培养的报道还不多。

本研究力求探索紫叶矮樱组织培养快速繁殖的有效途径。采用正交实验设计并结合统计分析,以山西农业大学经济林基地紫叶矮樱的带芽茎段为外植体,着重研究其离体培养的最佳培养基配方及培养程序,旨在建立紫叶矮樱快速繁殖技术体系,以期为低成本、高效率的工厂化育苗提供理论和技术依据。研究结果如下:

初代培养,外植体消毒采用5%NaClO处理20min,效果最好。3月下旬到4月中旬为最适取材季节。初代培养1/2MS+6-BA1.5mg/L+NAA0.05mg/L为最佳组合培养基配方。

继代培养,最佳培养基为1/2MS+6-BA0.5mg/L+NAA0.1mg/L,或者1/2MS+6-BA1.0

mg/L+NAA0.2mg/L,30d后其增殖率分别达到4.053.7

生根培养最佳培养基为1/2MS+IBA1.0mg/L+IAA1.0mg/L,其次为1/2MS+NAA1.5mg/L.一周时1/2MS+IBA1.0mg/L+IAA1.0mg/L有根长出,20d后平均生根数为25.4条,生根率为48.6%94.7%

With the development of the city garden green project, demand for the foliage plant introduce have increased. Subsequently the study on the foliage species have been also gradually listed schedule of city green. Prunus×cistena is looked on as world famous foliage species. The report on its tissue culture is fewer.

The study mainly explored the effective approach of tissue culture and instant reproduce for purpleleaf drart Primula.In my experiment, Prunus×cistena with apical bud stem segment as explants was chosen in the plant nursery of Shanxi Agriculture University, to investigate the best incubation media recipe and culture program of its isolated culture using statistic analysis. Instant reproduce system of Prunus×cistena is established finally, which provide theoretical and technical foundation for factory nursery garden with Low cost and high efficiencyThe results as follows:

1.Explant is sterilized with 5% NaClO adding a droplet TWEEN80 for 20 minutes ,Which are regarded as the best effectiveness.It is the most suitable for selecting material from the middle ten.days of March to the middle ten.days of April For the frist greneration, generation,1/2MS+6-BA1.5mg/L+NAA0.05mg/L is selected as the best combination incubation media recipe.

2.For the second generation culture,the best incubation media is 1/2MS+6-BA0.5mg/L+NAA0.1mg/L

or 1/2MS+6-BA1.0mg/L+NAA0.2mg/L,The propagation rate could reach about 4.05 and 3.7.

3.For the rooting culture, the best incubation media is 1/2MS+IBA1.0mg/L+IAA1.0mg/L,the better is 1/2MS+NAA1.5mg/.Root outgrows for 1/2MS+IBA1.0mg/L+IAA1.0mg/L after a week. The .average root numbers are in the range of 2 to 5.4 after 20 days. The rate of root is 48.6%to 94.7%.
